Your tasks
• Conduct research in mouse models to investigate how inhibition of endocytosis in Purkinje cells affects information processing in the thalamo-cortical network 
• Perform experiments in cerebellar organotypic slice cultures using GCaMP6-based calcium imaging and chemogenetics
• Perform stereotactic surgeries and use chemogenetics to manipulate brain motor circuits
• Conduct biochemical assays, such as pull-down experiments and proteomic analyses
• Maintain accurate lab records and clearly present findings in meetings

Your profile
• Master’s degree in Neuroscience, Cell Biology, or Biochemistry (with very good to excellent grades)
• Experience with techniques such as rodent models of
neurodegeneration, primary cell culture, immunohistochemistry, microscopy, image analysis, and biochemical assays
• Strong motivation to advance research in neurodegenerative diseases
• Proficiency in English
• Excellent organizational skills, a proactive attitude, enthusiasm, and a willingness to learn

The laboratory of Prof. Natalia Kononenko at the University of Cologne (Medical Faculty & CECAD Excellence Cluster) is looking for a highly motivated PhD candidate to investigate the role of the endocytosis in Purkinje cells and the impact of its deletion on brain-wide motor circuits (see Tolve et
al., 2024, 10.1101/2024.01.27.577372). This project is part of CRC1451 and involves advanced mouse models, rabies tracing, chemogenetics, multi-omics, super-resolution microscopy, miniscope Ca2+ imaging, offering an excellent
opportunity to engage in cutting-edge neurobiology research.

Im Sonderforschungsbereich (SFB) 1451 „Schlüsselmechanismen normaler und krankheitsbedingt gestörter motorischer Kontrolle" erforscht ein internationales, multidisziplinäres Team die genetischen Faktoren und zellulären, synaptischen und neuronalen Prozesse, die der motorischen Kontrolle bei Tieren und Menschen sowohl im gesunden Zustand als auch bei neuropsychiatrischen Erkrankungen zugrunde liegen.
Ziel ist es, allgemeine Mechanismen der motorischen Kontrolle, über eine einzelne Spezies oder eine spezifische Pathologie hinaus, abzuleiten sowie deren Veränderungen über die Lebensspanne und deren Störungen zu charakterisieren.
Hierfür kooperieren Wissenschaftlerinnen und Wissenschaftler der Medizinischen Fakultät und der Mathematisch-Naturwissenschaftlichen Fakultät der Universität zu Köln, der Uniklinik Köln, des Forschungszentrums Jülich, der Max-Planck-Institute für Stoffwechselforschung und Biologie des Alterns in Köln, der Goethe-Universität Frankfurt, der Universität Münster und der Hebrew University of Jerusalem. Das Projekt C04 des SFB 1451 befasst sich mit Gangstörungen bei der Alzheimer- (AD) und der Parkinson-Erkrankung (PD). Hierbei kommen bereits etablierte neuropsychologische Assessments, quantitative Ganganalysen sowie modernste klinisch/wissenschaftliche Methoden wie die tiefe Hirnstimulation (DBS) und Tau-PET-Bildgebung zur Anwendung. Die Forschungsarbeiten im Projekt C04 des SFB 1451 explorieren die Hypothese, dass die Dysfunktion eines gemeinsamen subkortikalen und kortikalen Schaltkreises den komplexen Gangstörungen bei AD und PD zugrunde liegt, was zu neuen Erkenntnissen über die Interaktion zwischen den motorischen und
kognitiven Systemen beim Gehen führen wird. Der/die erfolgreichen Kandidatinnen/Kandidat werden an der Vorbereitung und Durchführung der Untersuchungen in der Klinik für Neurologie, insbesondere von Verhaltensexperimenten an neurologischen Patientinnen und Patienten mit der Alzheimer- (AD) und der Parkinson-Erkrankung, sowie an der Analyse und Veröffentlichung von neu gewonnenen und bereits vorhandenen Datensätzen beteiligt sein. Die Stelle bietet hervorragende Möglichkeiten, insbesondere im integrierten Graduiertenkolleg des SFB.
Abgeschlossenes Hochschulstudium (Master) aus den Bereichen Psychologie, Neurowissenschaften, Ingenieurwesen, Physik, Biologie oder einem verwandten Gebiet
Sichere Englischkenntnisse zur Präsentation von wissenschaftlichen Ergebnissen und zum Erstellen wissenschaftlicher Publikationen
Idealerweise Erfahrung mit Programmiersprachen und Neuroimaging-Softwarepaketen
Erfahrung in der Durchführung statistischer Analysen
Motivation, mit Patientinnen und Patienten mit neurodegenerativen Erkrankungen zu arbeiten

Your profile

Your future in detail
The Ising lab is located in the CECAD Research building and as Junior Research Group part of the SFB 1403 on “Cell Death in Immunity, Inflammation and Disease" at the University of Cologne. We are investigating the role of cell death and senescence as contributors to the neuroinflammatory response in Alzheimer’s disease and other neurodegenerative diseases collectively termed tauopathies. Our overarching goal is to identify underlying molecular mechanisms in tauopathies to ultimately contribute to the development of new therapeutic strategies. We are combining primary cell culture work with mouse models, which we analyze with a multitude of techniques including but not limited to Western Blot, flow cytometry, ELISA, immunohistochemistry, RNAseq, proteomics and behavior tests. The main focus for this position will be the role of microglial senescence in tauopathies.
